Cloning and Expression Analysis of <i>AzanOBP7</i>, a Minus-C Odorant-Binding Protein Gene, from <i>Agrilus zanthoxylumi</i> (Coleoptera: Buprestidae)1

Odorant-binding proteins (OBPs) play an important role in specific recognition, binding, and transportation of odorants. In this study, the full-length complementary DNA (cDNA) sequence AzanOBP7, a Minus-C OBP gene, from Agrilus zanthoxylumi Li Ming Lou (Coleoptera: Buprestidae) was cloned by rapid amplification cDNA ends based on transcriptome data. The bioinformatic analysis showed that AzanOBP7 contains 450-bp open reading frame encoding 149-residue polypeptide, with molecular mass 17.176 kDa. It predicted to be nontransmembrane protein 18–amino acid signal peptide at N terminus. three-dimensional structure AphadFold2 possesses seven α helices two disulfide bridges. multiple alignment phylogenetic tree revealed reached highest identity (94.70%) mali Matsumura AmalOBP11; they also were closely aligned clade. Quantitative real-time polymerase chain reaction exhibited expression level abdomen adult females. thorax, males significantly higher than other aged (P < 0.01). Our study offers theoretical foundation for further functional characteristics A. OBPs.
